In mid-October 2026, following a turbulent nine-month stretch that saw Corvus Pharmaceuticals, Inc. (NASDAQ: CRVS) surge to an all-time high of $26.95 on January 22, 2026, execute a $201.2 million gross ($189.4 million net) follow-on public offering at $22.15 per share, and subsequently retrace over 55% into the $9.84 range amid broader small-cap biotech multiple compression, prominent independent target and mechanism researcher @HOThomasWPhelps (“In Honour Of Thomas W Phelps”) published a widely circulated statement addressing inbound shareholder pressure over whether Corvus should pursue an immediate corporate partnership:
“I am not a trader, I do research on targets, MOA, and drugs in development that interest me. If the potential is very high I may invest accordingly.
During periods of turmoil in biotech like now I get many email, DMs, PMs regarding the company I do most of my research on $CRVS. These messages have precipitated this post and my thoughts in one location.
My focus on Corvus and ITK inhibition is due to soquelitinib novel MOA that can be a paradigm shift in oncology and I&I. It stems from what I believe may lead to a drug that can be best in class from a combo therapy w/ ICB-treatment i.e., anti-PD-1/anti-CTLA4 to autoimmune disease where I believe soquelitinib could be best in disease for atopic dermatitis, asthma and maybe several others.
I do not trade Corvus or know what it’s price will do. Rather I just own shares in the company to have as much ownership of soquelitinib and its novel MOA as I can. I want to own soquelitinib. I recognise day to day price can be volatile, and over the last few months this has been more accute, this is not Corvus-centric, but biotech in general. Ironically these are the periods when I get the most inbounds from others. I have no advice.
I am a long-term investor and add and support Corvus and management best I can by being an anchor shareholder w/ large stable/growing shareholding.
A question I have gotten several times over the past week is regarding if Corvus should do a partnership or deal. In a 2 words, ‘not now.’ To me this is short-term thinking designed to pop the share price so someone can exit. Personally, I do not want a deal at this point and at this price level, that is short-term benefit as I said and would be giving huge potential future shareholder returns away that could be substantially larger.
Based on soquelitinib unique novel oral MOA, data to-date, trials ongoing (PTCL, ALPS, atopic dermatitis, HS) or about to start (asthma), cash position ($215M), pipeline w/ next-gen ITK inhibitors and target potential I think maximum shareholder returns/value will be found by waiting till next year, after several key data readouts and data has rolled in, before the company takes action on any partnership or transaction. I think SQL can be best in disease for atopic dermatitis and asthma, and maybe a few others.
I look forward to the data and trials that will initiate and readout over the next months and quarters through 2027 and to continue to watch Miller and team develop soquelitinib and other ITK inhibitors.“
— @HOThomasWPhelps, October 2026 (CRVS at $9.84)
The Phelps thesis sits at the intersection of deep T-cell receptor (TCR) enzymology and high-stakes biopharma capital allocation. Founded and led by Dr. Richard A. Miller, M.D.—the co-founder of IDEC Pharmaceuticals (rituximab / Rituxan) and former President, CEO, and co-founder of Pharmacyclics (ibrutinib / Imbruvica)—Corvus is attempting to repeat the covalent kinase playbook of ibrutinib in T lymphocytes by irreversibly targeting Cysteine-442 (Cys442) on Interleukin-2-inducible T-cell kinase (ITK) with its lead oral candidate, soquelitinib (formerly CPI-818).
To evaluate whether Phelps’s conviction is warranted by primary empirical evidence—or whether retail and institutional bulls are overlooking critical statistical, pharmacokinetic, and financial hazards—we execute an 8-phase “Panning for Gold” forensic investigation structured around two explicit, testable questions:
- The Descriptive (Mechanistic & Clinical) Question: Does covalent inhibition of ITK at Cys442 by soquelitinib genuinely achieve a selective “Th1-skewing and Treg-sparing” immune rebalance across both oncology (Peripheral T-Cell Lymphoma [PTCL] and immune checkpoint blockade [ICB] combinations) and Immunology & Inflammation (Atopic Dermatitis [AD], Asthma, Hidradenitis Suppurativa [HS], and Autoimmune Lymphoproliferative Syndrome [ALPS]), and can an oral ITK inhibitor realistically achieve “best-in-disease” efficacy against entrenched biologics (
dupilumab, lebrikizumab, amlitelimab, tezepelumab) and emerging oral targeted protein degraders (KT-621STAT6 degrader)? - The Prescriptive (Strategic & Financial) Question: With Corvus trading at $9.84 per share (~$909.2 million basic-plus-prefunded market cap, ~$694.0 million enterprise value, and $215.23 million in cash and marketable securities as of June 30, 2026), is “Not Now” the optimal game-theoretic strategy for partnerships, or does solo execution across five concurrent clinical programs expose shareholders to an unhedged Phase 2 regression cliff and a 2027–2028 Phase 3 capital wall?
Falsifiability Criteria
- What would falsify our skeptical adjustments (and validate the Blue-Sky Bull Thesis)? If Corvus’s 200-patient Phase 2 SIERRA-1 trial (
NCT07441395) in moderate-to-severe Atopic Dermatitis delivers a Week 12 placebo-adjusted EASI-75 delta ≥ 38% (absolute EASI-75 ≥ 55%–60%) with preserved 90-day off-drug remission across all three dosing arms (including 400 mg QD) and zero opportunistic viral/EBV signals, AND the Phase 3 PTCL trial (NCT06561048) clears its early 2027 interim futility analysis with a hazard ratio (HR) ≤ 0.60 for progression-free survival (PFS). - What would falsify the Phelps Bull Thesis? If multicenter Phase 2 dispersion in
SIERRA-1compresses the placebo-adjusted EASI-75 delta to < 20% (below dupilumab, upadacitinib, and Kymera’s oral STAT6 degraderKT-621), OR if the 400 mg once-daily (QD) arm exhibits paradoxical loss of Th1/Treg selectivity due to peak plasma (Cmax) spillover onto Resting Lymphocyte Kinase (RLK / TXK), OR if cash burn forces Corvus to tap its doubled $200.0 million Jefferies ATM facility at depressed share prices prior to securing a Tier-1 pharmaceutical partner.

1. Primary Source Excavation: Capital Structure, Insider Ownership & The 2-Column Claim Schism Matrix
To anchor our investigation in canonical legal and regulatory disclosures, we audited Corvus Pharmaceuticals’ Form 10-Q for the quarterly period ended June 30, 2026 (filed August 6, 2026), 2025 Annual Report on Form 10-K (filed March 12, 2026), 2026 Definitive Proxy Statement on Schedule 14A (filed April 24, 2026), and Form 8-K Current Report (filed May 14, 2026).
True Diluted Share Count & Enterprise Value at $9.84
Financial aggregator portals frequently underreport Corvus’s market capitalization by counting only basic common shares (84,120,944 shares as of August 6, 2026) while ignoring penny pre-funded warrants and deeply in-the-money employee stock options:
- Issued and Outstanding Common Stock: 84,120,944 shares ($0.0001 par value).
- Outstanding Pre-Funded Warrants ($0.0001 Exercise Price): 8,275,913 shares (issued in the May 2024 private placement and May 2025 common warrant exercise; included directly in Corvus’s GAAP basic EPS denominator of 92,368,013 weighted-average shares in Q2 2026). Per the 2026 Proxy Statement, 100% of these pre-funded warrants are held by two institutional healthcare funds:
- Sirenia Capital Management LP (Alex Silverstein): 6,831,828 pre-funded warrants (7.5% beneficial ownership).
- OrbiMed Private Investments V, LP (Peter A. Thompson, M.D., Board Member): 1,444,085 pre-funded warrants plus 7,165,006 common shares (8,609,091 total shares, 10.1% beneficial ownership).
- GAAP Basic + Pre-Funded Share Count: 92,396,857 shares (implying a $909.19 million market capitalization at $9.84/share).
- In-the-Money Stock Options: 13,253,806 options outstanding as of June 30, 2026, with a weighted-average exercise price (WAEP) of $4.90 per share (plus 6,431,034 shares available for future grant and 400,000 ESPP shares). Under the treasury stock method at a $9.84 share price, exercising these 13.25 million options generates $64.94 million in cash proceeds (repurchasing 6,600,000 shares at $9.84), adding 6,653,806 net dilutive shares.
- Fully Diluted Treasury-Method Share Count: 99,050,663 shares, equating to a Fully Diluted Market Capitalization of $974.66 million at $9.84/share.
- Cash, Cash Equivalents & Marketable Securities: $215.23 million as of June 30, 2026 ($18.905M in cash and cash equivalents + $170.234M in U.S. Treasury securities + $26.089M in U.S. government agency securities), bolstered by the January 23, 2026 follow-on offering of 9,085,778 shares at $22.15 per share ($189.407 million in net proceeds).
- Operating Burn Trajectory: In Q2 2026, R&D expenses jumped 102.8% year-over-year to $15.96 million (up from $7.87M in Q2 2025) and G&A rose to $3.33 million, bringing quarterly operating expenses to $19.30 million (6-month operating loss of $34.17 million; 6-month operating cash outflow of $25.77 million, plus $5.00 million invested in China partner Angel Pharmaceuticals Co. Ltd.). Factoring in estimated Q3 2026 net cash burn of
$15M–$17M, Corvus enters mid-October 2026 with approximately $198M–$200M in net cash ($2.00 per diluted share).

Insider Alignment & Governance Upgrading
To Phelps’s credit, insider alignment at Corvus is unusually high for a clinical-stage biotech:
- Dr. Richard A. Miller, M.D. (CEO) & Dr. Sandra J. Horning, M.D. (Former Chief Medical Officer of Genentech/Roche): Beneficially own 5,815,643 shares (6.6%), including 2,324,810 common shares held directly in the Miller Horning Family Trust and 3,490,833 exercisable options.
- All Executive Officers and Directors as a Group (10 Persons): Control 18,094,476 shares (19.7%).
- Strategic Board Addition (April 23, 2026): As disclosed in Corvus’s April 23, 2026 Form 8-K, Corvus appointed Dr. Andrew C. Chan, M.D., Ph.D.—former Senior Vice President of Research Biology at Genentech, and one of the pioneering immunologists who originally characterized ZAP-70 and proximal TCR tyrosine kinase cascades—to its Board of Directors.
The 2-Column Claim Schism Matrix: Bull Thesis vs. Regulatory Disclosures
Following Phase 2 of our runbook, we cross-examined the public claims circulating in retail/institutional bull commentary against buried disclosures in Corvus’s SEC filings, NIH clinical trial exclusions, and peer-reviewed enzymology data:
| PUBLIC BULL NARRATIVE (@HOThomasWPhelps / Street) | BURIED SEC FILING, PROTOCOL & ENZYMOLOGY REALITY |
|---|---|
| 1. “Combo therapy w/ ICB-treatment i.e., anti-PD-1 / anti-CTLA4 can lead to a best-in-class drug in oncology.” | In Q2 2026 Form 10-Q (p. 30), Corvus’s clinical ipilimumab + nivolumab RCC combo trial used CIFORADENANT (A2AR antagonist), which hit a non-significant 34% vs 32% deep response rate at ESMO Oct 2025 (“No further trials are planned”). Soquelitinib + ICB has ONLY been tested in mice. |
| 2. “Cohort 4 AD showed 75% EASI-75 vs 20% placebo (p=0.035), proving best-in-disease efficacy across systemic-naive and refractory patients.” | Form 10-Q (p. 25-28) reveals Cohort 4 enrolled N=12 active vs N=12 placebo (2 placebo missed Day 56). Randomization was skewed: 58.3% (7/12) of placebo had failed prior systemics vs 41.7% (5/12) on active drug, depressing the placebo arm. |
| 3. “Phase 1/1b PTCL data in Blood showed 37.5% ORR (25% CR) and 28.1-month median OS at 200 mg BID.” | Blood (Aug 2026, PMID: 42640855) & 10-Q (p. 23-24) admit that across all N=36 patients treated at 200 mg BID, all 9 responses occurred in the N=24 subgroup with 1-3 prior lines & adequate ALC; patients with >3 prior lines had 0% ORR (0/12). |
| 4. “Specificity of soquelitinib for ITK confers safety without immunosuppression.” | NIAID Phase 2a ALPS protocol (NCT06730126) explicitly excludes active EBV (>300 copies/mL), history of EBV lymphoma, and moderate/strong CYP3A4 or P-gp inhibitors, monitoring Grade 3/4 viral/mycobacterial/fungal infections for 360 days. |
| 5. “$215M cash position means Corvus does not need any deal or financing until late 2027.” | On March 13, 2026 (Form 10-Q Note 10, p. 16), immediately after raising $189.4M net at $22.15, Corvus amended its Jefferies ATM agreement to DOUBLE its equity shelf capacity from $100M to $200M, preparing for Phase 3 I&I capital needs. |
2. Semantic Precision & Biophysical Audit: The Covalent ITK vs. RLK “Goldilocks Window”
In Phase 3 of our methodology, we apply the Substitution Test to three ambiguous phrases in @HOThomasWPhelps’s post:
- “Selective ITK Inhibition / Novel MOA”
- “Combo Therapy w/ ICB-Treatment (anti-PD-1 / anti-CTLA-4)”
- “Best in Disease for Atopic Dermatitis and Asthma”
The Substitution Test on “Selective ITK Inhibition”
Why have pharmaceutical giants including Pfizer (PF-06465469), Bristol Myers Squibb (BMS-509744), Genentech/Roche (GNE-4997 / GNE-9822), GlaxoSmithKline (inhaled ITK series), and Principia Biopharma/Sanofi (PRN694) spent 15 years attempting to drugging ITK for asthma, psoriasis, and autoimmune disease without a single FDA approval?
The answer lies in the TEC-family kinase homology problem. In human T cells, two closely related TEC-family non-receptor tyrosine kinases—ITK (Interleukin-2-inducible T-cell kinase) and RLK (Resting Lymphocyte Kinase, also known as TXK)—operate downstream of the T-cell receptor (TCR) and ZAP-70 to phosphorylate PLCγ1, triggering calcium flux (NFAT), NF-κB, ERK, and mTOR signaling:

In classic murine genetic studies (Itk-/- single-knockout vs. Itk-/- Rlk-/- double-knockout mice), deleting Itk alone impairs TCR signal strength just enough to block Th2 (GATA3) and Th17 (RORγt) differentiation while shifting naive CD4+ T cells toward Th1 (T-bet) and Regulatory T cells (Foxp3+ Tregs), because RLK/TXK remains intact to rescue Th1 and CD8+ cytotoxic T lymphocyte (CTL) function. However, when both ITK and RLK are inhibited (Itk-/- Rlk-/- double knockout or dual ITK/RLK inhibitor PRN694), PLCγ1 phosphorylation collapses completely, paralyzing CD8+ granzyme B, IL-2, and IFN-γ production and causing global T-cell immunosuppression.
Mathematical Derivation of Soquelitinib’s Covalent Kinetic Window
According to Corvus’s foundational peer-reviewed characterization in npj Drug Discovery (December 2024, PMID: 42380486), soquelitinib uses an aminothiazole scaffold that forms two hydrogen bonds with Met438 in the ITK hinge region, a hydrophobic contact between its 2-methyl group and the Phe435 gatekeeper residue, and a methyldiazepane linker that positions an acrylamide warhead to form an irreversible covalent carbon-sulfur bond with Cys442 (confirmed across 247 chymotryptic peptide fragments via LC-MS/MS).
However, all five TEC-family kinases (ITK, RLK/TXK, BTK, BMX, TEC) and six other human kinases (JAK3, EGFR, ERBB2, ERBB4, BLK, MKK7) possess a homologous cysteine residue at the exact Cys442 position. Because covalent binding is a two-step irreversible reaction (E + I ⇄ E·I → E-I*), true biological selectivity in living T cells is governed not by equilibrium binding (Kd), but by the competition between the pseudo-first-order covalent inactivation rate (k_obs,i) and the endogenous protein resynthesis/turnover rate (k_deg,i = ln(2) / t_1/2,i) at free plasma/intracellular concentration [I]_free:
$$k_{\text{obs}, i}\left([I]{\text{free}}\right) = \frac{k{\text{inact}, i} \cdot [I]{\text{free}}}{K{I, i} + [I]{\text{free}}} \approx \left(\frac{k{\text{inact}, i}}{K_{I, i}}\right) \cdot [I]{\text{free}} \quad \text{for } [I]{\text{free}} \ll K_{I, i}$$
At steady state under twice-daily (BID) oral dosing, the fractional covalent target occupancy O_i([I]_free) for kinase i ∈ {ITK, RLK} is:
$$O_i\left([I]{\text{free}}\right) = \frac{k{\text{obs}, i}\left([I]{\text{free}}\right)}{k{\text{obs}, i}\left([I]{\text{free}}\right) + k{\text{deg}, i}} = \frac{1}{1 + \dfrac{k_{\text{deg}, i}}{\left(k_{\text{inact}, i} / K_{I, i}\right) \cdot [I]_{\text{free}}}}$$
We define the Th1-Skewing Selectivity Window (ΔO_Th1) as the differential steady-state occupancy between ITK and RLK:
$$\Delta O_{\text{Th1}}\left([I]{\text{free}}\right) = O{\text{ITK}}\left([I]{\text{free}}\right) - O{\text{RLK}}\left([I]_{\text{free}}\right)$$
From the enzymatic data published in npj Drug Discovery (Hsu et al., 2024):
- ITK Binding & Inactivation:
Kd,ITK = 6.5 nM; enzymatic inactivationIC50,ITK = 0.5 nM; second-order covalent potencykinact,ITK / KI,ITK = 1.71 µM⁻¹s⁻¹(suppressing Jurkat TCR-stimulated IL-2 secretion with cellularIC50 = 136 nM). - RLK (TXK) Inactivation: Enzymatic inactivation
IC50,RLK = 261 nM; second-order covalent potencykinact,RLK / KI,RLK = 0.0149 µM⁻¹s⁻¹(115-fold kinetic selectivity bykinact/KI, and 522-fold selectivity by enzymaticIC50). - Structurally Similar Non-Selective Analog (Fig. 4 Control):
IC50,ITK = 0.6 nMvs.IC50,RLK = 2.2 nM(only 3.7-fold separation), which severely suppressed CD8+ CTL IFN-γ, IL-2, and granzyme B.
What happens when we substitute different intracellular concentrations [I]_free into our occupancy model and compare them against the published cellular curves in Figures 2e, 3b, 4, and 7b of npj Drug Discovery?
- Definition A (The 0.1–1.0 µM “Goldilocks Regime” — Achieved by 200 mg BID): At
[I]_free ≈ 0.1–0.5 µM, ITK occupancy exceeds 85%–95% while RLK inactivation remains minimal. In human CD4+ T cells (Fig. 3b), the intracellular ratio ofIFN-γ+toIL-4+cells increases by > 2-fold at 1.0 µM, and exhausted OT-1 CD8+ T cells (Fig. 7b) downregulateLAG3,TIGIT, andPD-1while restoring granzyme B and IFN-γ secretion. - Definition B (The ≥ 3–10 µM “Spillover & Arrest Regime”): Because covalent inactivation is irreversible, excessive drug concentrations (
[I]_free → 3–10 µM) force covalent modification of RLK (IC50 = 261 nM) and off-target kinases (ZAP-70is inhibited 41% at 1.0 µM; 8 kinome enzymes are inhibited ≥ 65% at 1.0 µM). As the authors explicitly admit in npj Drug Discovery:- “At a concentration of 10 μM, soquelitinib completely abolished production of either IL-4 or IFNγ in naïve CD4+ T cells, possibly due to its antiproliferative effects at high concentrations (Fig. 2e).”
- “Varying concentrations of soquelitinib restored these functions [granzyme B and IFNγ in exhausted OT-1 CD8 T cells]… except at the highest 10 μM concentration tested.”
Forensic Takeaway from the Substitution Test: Soquelitinib is not a monotonic “more drug = better efficacy” inhibitor. It operates on a strict bell-shaped immunomodulatory curve. If systemic or tissue Cmax overshoots the 115-fold ITK/RLK selectivity buffer, the drug transitions from an immune-rebalancing Th1/Treg modulator into a pan-T-cell antiproliferative immunosuppressant.

Phase 4 & 5 Forensic Deconstruction: Four Critical Adjustments to Cohort 4
While a 75.0% EASI-75 rate (9/12) vs. 20.0% (2/10) placebo at Week 8—accompanied by zero disease flares during a 30-to-90-day drug-free follow-up period and identical adverse event rates between drug and placebo (41.7% vs. 41.7%, all Grade 1–2)—is an undeniably promising Phase 1 signal, our statistical and causal audit uncovers four crucial caveats that must be priced into Phase 2 SIERRA-1:
- Exact Clopper-Pearson Binomial Confidence Intervals:
For a binomial sample of
n = 12withk = 9successes (75.0%), the exact two-sided 95% Clopper-Pearson confidence limits[p_L, p_U]satisfy:
$$\sum_{j=k}^{n} \binom{n}{j} p_L^j (1 - p_L)^{n-j} = 0.025, \qquad \sum_{j=0}^{k} \binom{n}{j} p_U^j (1 - p_U)^{n-j} = 0.025$$
Solving for n = 12, k = 9 yields a 95% CI of [42.8%, 94.5%], while the placebo group (k = 2, n = 10, 20.0%) spans [2.5%, 55.6%]. On the FDA’s preferred registrational co-primary endpoint of vIGA 0/1 (4/12 = 33.3%), the exact 95% CI spans [9.9%, 65.1%]. Simply put, a single patient shifting from responder to non-responder in a 12-patient cohort moves the headline rate by 8.3 percentage points.

The Hidden Prior-Systemic Randomization Skew in Cohort 4: Look closely at pages 25–28 of the Q2 2026 Form 10-Q. In Cohort 4 (
N = 24), exactly 12 patients (50%) had received prior systemic therapy (such asdupilumabor oral JAK inhibitors). However, because Cohort 4 did not stratify randomization by prior systemic exposure, 5 of the 12 soquelitinib patients (41.7%) were systemic-experienced, whereas 7 of the 12 placebo patients (58.3%) were systemic-experienced! As Corvus’s 10-Q explicitly states:“Also, prior systemic therapy experienced patients appeared to have worse disease. The two placebo patients who achieved EASI 75 had not received prior systemic therapy. None of the seven placebo patients who had received prior systemic therapy achieved EASI 75 or EASI 50; whereas, three of the five soquelitinib treated patients who received prior systemic therapy achieved EASI 75.”
Notice the mathematical consequence of this baseline imbalance:
- In the systemic-naive stratum of Cohort 4, placebo achieved EASI-75 in 2/3 (66.7%) or 2/5 (40.0%) of evaluable patients (and active achieved
6/7 = 85.7%). - Because the placebo arm was disproportionately loaded with systemic-refractory patients (
7/12 = 58.3%) who had a 0% (0/7) placebo response, the blended Cohort 4 placebo EASI-75 rate was artificially held down to 20.0% (2/10), even as mean EASI reduction in the placebo group reached a remarkably high 40.0% at Day 56 (and 42.1% in Cohort 3)!
- In the systemic-naive stratum of Cohort 4, placebo achieved EASI-75 in 2/3 (66.7%) or 2/5 (40.0%) of evaluable patients (and active achieved

4-Way Causal Disentanglement of the 30-to-90-Day “Drug-Free Remission”: Why did Cohort 3 and 4 patients maintain EASI reductions for 30 to 90 days after stopping pills without rebound flares?
- Direct Mechanistic Pathway (Validated by scRNA-seq & Flow Cytometry): In the May 14, 2026 Form 8-K (
Figures 4a–6b), Corvus demonstrated that soquelitinib downregulatesRORγt(master Th17 transcription factor) while upregulatingBACH2(which stabilizes theFoxp3promoter), driving a direct Th17-to-Treg lineage switch that expands circulatingCD4+ Foxp3+ CD25hipersistent Tregs both at Day 28 and 30 days post-treatment, alongside increasedSOCS3and reducedJAK1/STAT6in circulating Th2 cells. - Mutual Feedback Loop (The Itch-Scratch-Alarmin Cycle): In atopic dermatitis, mechanical scratching damages keratinocytes, releasing epithelial alarmins (
TSLP,IL-33,IL-25) that perpetually re-stimulate cutaneous Th2/ILC2 cells. Once an 8-week course suppressesIL-4,IL-5,IL-13, andTARC(CCL17) enough to restore epidermal barrier integrity and halt scratching, even placebo responders who maintain intensive emollient skincare can remain flare-free for 4–8 weeks before subclinical re-sensitization triggers a relapse.

Benchmarking “Best-in-Disease” Against the 2026–2027 AD Competitive Field: For soquelitinib to be crowned “best in disease” in Phase 2
SIERRA-1(Week 12 readout expected mid-2027), it does not merely compete against legacy biologics; it faces a fiercely crowded oral and long-acting biologic arena:
2026-2027 MODERATE-TO-SEVERE ATOPIC DERMATITIS COMPETITIVE BENCHMARK
| Drug / Sponsor | Modality & Target | Key Efficacy Metric | Strategic Advantage / Liability |
|---|---|---|---|
| Soquelitinib (CPI-818) Corvus ($CRVS) | Oral Covalent ITK Inhibitor (Cys442) | P1 Coh 4 (8-Wk, N=12): 75% EASI-75 (55% Δ) | Oral BID/QD; Th17->Treg switch; Unproven in N=200 12-Wk P2 |
| KT-621 Kymera / Sanofi | Oral STAT6 Targeted Protein Degrader | P1b BroADen (4-Wk): 63% Mean EASI Reduct. | Once-daily oral; 93.6% skin STAT6 knockdown; P2b mid-2027 |
| Upadacitinib (Rinvoq) AbbVie | Oral JAK1 Selective Inhibitor (15/30 mg) | P3 Measure Up (16-Wk): 70-80% EASI-75 (~55%Δ) | Gold-standard oral efficacy, BUT carries FDA Black Box Warn |
| Dupilumab (Dupixent) Regeneron / Sanofi | SubQ Anti-IL-4Rα mAb (Q2W Biologic) | P3 SOLO 1/2 (16-Wk): 44-51% EASI-75 (~35%Δ) | Clean multi-year safety; ~$14B+ entrenched 1L biologic standard |
| Amlitelimab Sanofi | SubQ Anti-OX40L mAb (Non-depleting) | P3 COAST/SHORE (24W): ~47-50% EASI-75 | Q12W maintenance dosing; targets T-cell memory / Treg |
If soquelitinib replicates a 50%–60% absolute EASI-75 (30%–35% placebo-adjusted delta) at Week 12 in SIERRA-1 with zero JAK-style boxed warning liabilities (no thrombosis, MACE, acne, or herpes zoster reactivation) and 90-day off-drug durability, it would genuinely rival Kymera/Sanofi’s oral STAT6 degrader KT-621 for the title of best-in-class oral immunology therapy. Conversely, if multicenter placebo rates remain near 35%–40% (as seen on mean EASI in Cohorts 3 and 4) while active EASI-75 regresses toward 40%–45%, its placebo-adjusted delta would compress into the 15%–20% range.
4. Auditing the Oncology Thesis: The PTCL Subgroup Filter & The ICB Combo Myth
1. The Blinding Test on Phase 1/1b PTCL (NCT03952078 / Blood August 2026)
On August 25, 2026, Dr. Richard Miller and colleagues published the final peer-reviewed results of the Phase 1/1b trial of soquelitinib in relapsed/refractory (R/R) T-cell lymphomas in Blood (PMID: 42640855).
In investor presentations and social media summaries, bulls highlight three headline figures from the 200 mg BID recommended dose cohort:
- Objective Response Rate (ORR): 37.5% (9/24), including 6 Complete Responses (25.0% CR) and 3 Partial Responses (12.5% PR), with several CRs exceeding 2 years.
- Median Progression-Free Survival (PFS): 6.2 months (30% PFS at 18 months).
- Median Overall Survival (OS): 28.1 months (67% OS at 24 months), compared to historical salvage benchmarks of 3–4 months median PFS and 6–12 months median OS.
When we apply the Phase 5 Blinding & Denominator Audit to the raw numbers in the Blood paper and page 23–24 of the Q2 2026 Form 10-Q, we uncover the exact post-hoc stratification that generated those numbers:
- The trial enrolled 75 patients total across dose escalation (100, 200, 400, and 600 mg BID;
n = 27) and dose expansion (n = 48). - A total of
N = 36patients were treated at the selected 200 mg BID dose. - Across all 36 patients at 200 mg BID, there were 9 total responses (
9/36 = 25.0%unfiltered ORR;6/36 = 16.7%unfiltered CR). - All 9 responses occurred inside a retrospectively defined subset of
n = 24patients who met two criteria: (1) between 1 and 3 prior lines of systemic therapy, and (2) an adequate baseline peripheral blood absolute lymphocyte count (ALC). - In the remaining
n = 12patients at 200 mg BID who had received > 3 prior lines of therapy (or were severely lymphopenic), the objective response rate was 0.0% (0/12)!
Why did patients with > 3 prior lines or low ALC experience a 0% response rate? Because soquelitinib is not a direct cytotoxic chemotherapy (as confirmed in npj Drug Discovery Supplementary Fig. 4, where soquelitinib had zero direct cytotoxicity against lymphoma cell lines in vitro). Instead, soquelitinib relies on intact normal host CD8+ and Th1 effector T cells to infiltrate the tumor microenvironment and reject malignant Th2/TFH-polarized lymphoma cells! When a patient has been ravaged by 4 to 18 prior lines of lymphodepleting chemotherapy, their normal host T-cell compartment is too exhausted and depleted to mount an anti-tumor immune response.
Why This Actually Validates Dr. Miller’s Phase 3 Design (NCT06561048):
Rather than blindly marching all-comer R/R PTCL patients into Phase 3, Dr. Miller used this Phase 1 mechanistic insight to lock the inclusion criteria of the registrational 150-patient Phase 3 trial (NCT06561048, initiated October 2024) strictly to patients with ≥ 1 and ≤ 3 prior therapies, randomized 1:1 against physician’s choice of belinostat (historical ORR 25.8%, CR 10.8%, median PFS 1.6 months) or pralatrexate (historical ORR 27.0%, CR 11.0%, median PFS 3.5 months). By excluding the > 3 prior-line non-responders by protocol design, Corvus has substantially increased the probability that soquelitinib beats single-agent belinostat/pralatrexate on the primary endpoint of IRC-assessed PFS at the early 2027 interim futility analysis and November 2027 primary completion.
2. Correcting the ICB Combo Conflation
In his post, @HOThomasWPhelps writes that his focus on soquelitinib “stems from what I believe may lead to a drug that can be best in class from a combo therapy w/ ICB-treatment i.e., anti-PD-1/anti-CTLA4.”
Our primary-source audit of ClinicalTrials.gov and Corvus’s Q2 2026 Form 10-Q requires an immediate factual correction:
- Soquelitinib has never been tested in combination with anti-PD-1 or anti-CTLA-4 in humans. Every piece of data combining soquelitinib with anti-PD-1 + anti-CTLA-4 comes strictly from preclinical murine CT26 colon carcinoma models (
Fig. 6of npj Drug Discovery, 2024). - The clinical trial Corvus actually conducted in combination with
ipilimumab(anti-CTLA-4) andnivolumab(anti-PD-1) in first-line metastatic Renal Cell Carcinoma (RCC) used a completely different molecule: ciforadenant (CPI-444), an oral adenosine A2A receptor antagonist licensed from Vernalis (NCT03454451/ KCRC collaboration). - As disclosed on page 30 of the Q2 2026 Form 10-Q, ciforadenant + ipilimumab/nivolumab reported interim Phase 1b/2 results at ESMO in October 2025: it achieved a 34% deep response rate compared to the 32% historical benchmark for ipi/nivo alone (missing the pre-specified 50% relative improvement threshold of 48%), and Corvus disclosed: “No further trials are planned.”
While recent independent academic work in Cell Death Discovery (March 2026, PMID: 41792111) showed that ex vivo/in vivo ITK remodeling by soquelitinib reduces CAR-T exhaustion and boosts GZMB, TNF-α, and IFN-γ in anti-CD19 CAR-T therapy, any valuation attributed to human soquelitinib + ICB combination therapy today is purely preclinical option value.
5. The “Negative View” & Omission Interrogation: Graveyard Audits, Human EBV Genetics & The 400 mg QD Trap
Phase 6 of our runbook requires interrogating what company presentations and bull threads omit. We identify three structural biological and pharmacokinetic risks that every CRVS shareholder must monitor:
1. The 15-Year ITK Inhibitor Graveyard Audit
Why did prior pharma programs fail to translate murine ITK asthma/dermatitis efficacy into human drugs?
THE ITK INHIBITOR GRAVEYARD VS. SOQUELITINIB (SURVIVORSHIP AUDIT)
| Compound & Sponsor | Binding Mechanism | Biochemical Profile | Primary Cause of Failure |
|---|---|---|---|
| BMS-509744 Bristol Myers Squibb | Reversible ATPcompetitive | ITK IC50 = 19 nM Short residence time | Outcompeted by 1-5 mM intracellular ATP; poor PK |
| GNE-4997 / GNE-9822 Genentech / Roche | Reversible / Covalent indazole series | Potent ITK, moderate kinome selectivity | Preclinical toxicity & poor oral asthma therapeutic idx |
| PF-06465469 Pfizer | Covalent Cys442 acrylamide | kinact/Ki = 0.016 µM⁻¹s⁻¹ Equal potency on BTK | 107x weaker than SQL; equal BTK blockade -> off-target |
| GSK Inhaled ITK Series GlaxoSmithKline | Covalent Cys442 inhaled scaffold | kinact/Ki = 1.9 µM⁻¹s⁻¹ | Off-target BTK IC50 = 80 nM & lung irritation liabilities |
| PRN694 Principia / Sanofi | Covalent Dual ITK / RLK Inhibitor | ITK kinact/Ki = 4.7 RLK kinact/Ki = 0.46 | Dual RLK blockade crushed Th1/CD8+ IFN-γ & Granzyme B |
| Soquelitinib (CPI-818) Corvus ($CRVS) | Covalent Cys442 aminothiazole | ITK kinact/Ki = 1.71 RLK kinact/Ki = 0.0149 | SURVIVOR: 115x selective vs RLK; spares BTK & Th1/CD8+ |
Our graveyard audit confirms that Corvus solved the specific biochemical flaw (RLK and BTK cross-reactivity) that killed PRN694 and PF-06465469. However, solving off-target kinase cross-reactivity does not eliminate on-target human ITK biology.
2. On-Target Human Genetic Toxicology: The EBV & Mycobacterial Liability (OMIM #613011)
In specific-pathogen-free (SPF) laboratory cages, Itk-/- knockout mice appear healthy. In humans, however, homozygous loss-of-function mutations in the ITK gene cause a life-threatening autosomal recessive primary immunodeficiency (ITK Deficiency, OMIM #613011).
- As documented in the Journal of Experimental Medicine (2023, PMID: 36326697) and Clinical and Experimental Immunology (January 2026, PMID: 42132280), human ITK is non-redundantly required for invariant Natural Killer T (iNKT) cell ontogeny, CD8+ T-cell control of Epstein-Barr Virus (EBV), and IFN-γ immunity against Mycobacterium tuberculosis. Humans lacking functional ITK suffer from catastrophic EBV viremia, EBV-driven B-cell lymphoproliferative disorders, Hodgkin/non-Hodgkin lymphomas, and multifocal EBV-associated smooth muscle tumors.
- Look at what the NIH/NIAID investigators wrote into the ALPS-FAS clinical protocol (
NCT06730126): When Dr. V. Koneti Rao and the NIAID team designed the 360-day Phase 2a study of soquelitinib (200 mg BID) in Autoimmune Lymphoproliferative Syndrome (ClinicalTrials.gov NCT06730126), they inserted four strict safety guardrails that rarely appear in investor decks:- Exclusion Criterion 4: “History of EBV-associated lymphoma.”
- Exclusion Criterion 5: “Active EBV infection with EBV load > 300 copies/mL.”
- Exclusion Criterion 6: “Tuberculosis infection (active or latent) or undergoing tuberculosis treatment.”
- Secondary Endpoints 5 & 6: Formal 360-day tracking of “Grade 3 or 4 systemic infections” and “Clinically significant worsening of viral, mycobacterial, or fungal infection requiring new treatment.”
While short 4-to-12-week courses of soquelitinib in adult AD patients with established anti-EBV memory CTLs have shown zero viral reactivation to date, chronic multi-year ITK inhibition in pediatric or adolescent eczema and asthma patients (the exact age group undergoing primary EBV seroconversion / infectious mononucleosis) will face intense FDA scrutiny.
3. The SIERRA-1 400 mg QD Peak Exposure (Cmax) Trap & CYP3A4 / P-gp Drug-Drug Interactions
In Phase 2 SIERRA-1 (NCT07441395), Corvus is randomizing 200 patients across four arms (n ≈ 50 per arm) for 12 weeks:
- Cohort 1: Soquelitinib 200 mg once daily (QD)
- Cohort 2: Soquelitinib 200 mg twice daily (BID) (the validated Phase 1 Cohort 3/4 regimen)
- Cohort 3: Soquelitinib 400 mg once daily (QD)
- Placebo: Matching oral tablets twice daily
Why did Corvus add a 400 mg once-daily (QD) arm? For commercial convenience against once-daily oral rivals (KT-621, upadacitinib). However, because ITK is replenished by de novo protein synthesis (t_1/2 ≈ 12–24 hours) while RLK is spared only when peak free plasma concentrations remain well below IC50,RLK = 261 nM, administering 400 mg in a single bolus (QD) rather than splitting it into 200 mg every 12 hours (BID) produces a significantly higher peak concentration (Cmax) followed by a deeper 24-hour trough (Cmin):
- During the first 2–4 hours post-dose, a 400 mg QD bolus risks pushing intracellular
[I]_freeinto the RLK covalent inactivation zone, blunting the very Th1/Treg switch that gives soquelitinib its mechanism! - Furthermore, as revealed in Exclusion Criteria 11 and 12 of the NIH ALPS protocol (
NCT06730126), soquelitinib is sensitive to moderate or strong CYP3A4 inhibitors/inducers and P-glycoprotein (P-gp) inhibitors (such as clarithromycin, itraconazole, ketoconazole, verapamil, or diltiazem). In a 200-patient multicenter trial, any concomitant CYP3A4/P-gp inhibition could elevate exposure into the pan-T-cell antiproliferative regime.
6. Auditing the Prescriptive Dilemma: Is “Not Now” Optimal Game Theory for Partnerships?
We now turn to @HOThomasWPhelps’s central prescriptive thesis:
“A question I have gotten several times over the past week is regarding if Corvus should do a partnership or deal. In a 2 words, ‘not now.’ … Based on soquelitinib unique novel oral MOA, data to-date, trials ongoing (PTCL, ALPS, atopic dermatitis, HS) or about to start (asthma), cash position ($215M), pipeline w/ next-gen ITK inhibitors and target potential I think maximum shareholder returns/value will be found by waiting till next year, after several key data readouts and data has rolled in, before the company takes action on any partnership or transaction.”
To test whether “Not Now” maximizes risk-adjusted shareholder value, we model Corvus’s cash burn trajectory and Phase 3 capital requirements from Q2 2026 through Q2 2028.
The Cash Burn & Phase 3 I&I Capital Wall Math
-
Current Cash & Projected Mid-2027 Balance:
- Starting Cash & Marketable Securities (June 30, 2026): $215.23 million.
- In Q2 2026, before the 200-patient
SIERRA-1AD trial, 25-patient Phase 1b HS trial (started September 17, 2026), and Phase 2 Asthma trial reached full enrollment velocity, quarterly operating expenses had already nearly doubled to $19.30 million/quarter ($15.96M R&D + $3.33M G&A), partially offset by ~$2.05M/quarter in Treasury interest income. - As all four company-sponsored clinical trials (
NCT06561048Phase 3 PTCL,NCT07441395Phase 2 AD,NCT07828860Phase 1b HS, and Phase 2 Asthma) plus CMC drug supply scale concurrently across 2H 2026 and 2027, quarterly operating expenses will rise to $24.0M–$28.0M per quarter (net cash burn of ~$22M–$25M/quarter or ~$90M–$100M/year). - Consequently, when
SIERRA-1reads out in June 2027, Corvus’s cash balance will have declined from $215.2M to approximately $120M–$130M—leaving roughly 4.5 to 5 quarters of runway (into Q2 2028, matching management’s exact 10-Q guidance).
-
The Cost of Phase 3 Immunology & Inflammation (AD + Asthma + HS):
- Unlike orphan oncology (where a single 150-patient PTCL trial costing ~$45M–$60M suffices for FDA registration), FDA registration for an oral systemic small molecule in Atopic Dermatitis requires two replicate global Phase 3 trials (
N = 600–800each) plus a 52-week long-term safety database of≥ 1,000–1,500exposed patients (costing $250M–$350M). - Adding Phase 3 programs in Asthma (
N > 1,200, $200M–$300M) and Hidradenitis Suppurativa ($150M–$200M) requires $600M–$850M in cumulative development capital. - This is why Corvus doubled its Jefferies ATM shelf from $100M to $200M on March 13, 2026 (Form 10-Q Note 10, p. 16): management knows that $215.2M funds Phase 2 proof-of-concept, not Phase 3 I&I registration.
- Unlike orphan oncology (where a single 150-patient PTCL trial costing ~$45M–$60M suffices for FDA registration), FDA registration for an oral systemic small molecule in Atopic Dermatitis requires two replicate global Phase 3 trials (
Expected Value Comparison Across Three Strategic Postures
Let p_win be the probability of a decisive Phase 2 SIERRA-1 + PTCL interim win in 1H 2027, p_mod be the probability of a moderate/mixed readout, and p_fail be the probability of clinical failure:
$$\text{E}\left[V_{\text{sh}}\right] = \frac{p_{\text{win}} \cdot EV_{\text{win}} + p_{\text{mod}} \cdot EV_{\text{mod}} + p_{\text{fail}} \cdot EV_{\text{fail}} + \text{Cash}{\text{2027}} + \text{Upfront}{\text{deal}}}{N_{\text{diluted}} + \Delta N_{\text{offering}}}$$
STRATEGIC PARTNERSHIP GAME-THEORY MATRIX (OCTOBER 2026 - MID-2027)
| Strategic Posture | Upside / Downside Mechanics | Forensic Assessment |
|---|---|---|
| Posture 1: Premature Full Buyout or Cheap Global Out-License Today at $9.84/sh | • Caps near-term stock at ~$14-$16 • Gives away 70-80% of AD/Asthma economics at a 55% discount to the Jan 2026 ($22.15) offering |
SUBOPTIMAL. @HOThomasWPhelps is 100% right that signing a fire-sale global deal at $9.84 after a macro biotech drawdown destroys long-term EV. |
| Posture 2: Pure Unhedged Solo Run Through Late 2027 (“Phelps Pure Solo”) | • Retains 100% ex-China upside into June 2027 SIERRA-1 readout • Enters mid-2027 with ~$120M cash and $200M Jefferies ATM overhang |
HIGH BINARY & DILUTION RISK. If SIERRA-1 is moderate (e.g., 18-22% EASI-75 Δ), Corvus has <12 mos cash and must tap its $200M ATM or negotiate from weakness |
| Posture 3: Staged Indication-Split or Ex-US / Asthma Deal After 1Q27 Interim Data | • Wait for Angel China AD Cohort 1 (Late 2026) & PTCL Interim (1Q27) • Out-license Asthma/Ex-US I&I for $150M-$250M upfront; keep PTCL/AD |
OPTIMAL RISK-ADJUSTED EV. Uses China 12-wk AD & PTCL interim to re-rate stock to $15-$18, then secures non-dilutive cash before SIERRA-1 binary. |
Where Phelps Is Right—and What He Misses: Phelps is unequivocally right that Corvus should reject any lowball buyout or global I&I licensing giveaway today at $9.84 per share (~$700M EV), especially when institutional investors just funded the company at $22.15 per share nine months ago and four value-inflection readouts sit within the next 3 to 9 months:
- Late 2026 (Q4 2026): Angel Pharmaceuticals (China) Phase 1b/2 AD Cohort 1 (12-week treatment data at 100 mg BID and 200 mg QD vs. placebo—providing our first glimpse of 12-week EASI kinetics prior to
SIERRA-1!). - December 2026 / 1H 2027: NIH/NIAID Phase 2a ALPS-FAS (
NCT06730126) Day 90 CT/PET-CT spleen and lymph node volume reduction readout. - Early 2027 (1Q 2027): Phase 3 R/R PTCL (
NCT06561048) Interim Futility Analysis on IRC-assessed PFS. - Mid-2027 (June 2027): Phase 2
SIERRA-1(NCT07441395) 200-patient, 12-week Atopic Dermatitis primary readout.
However, treating corporate strategy as a binary choice between “sell out cheap today” and “refuse all partnerships until late 2027” overlooks the reality that even a victorious Phase 2 SIERRA-1 readout in June 2027 will immediately require a $250M–$400M capital injection (via either a major Big Pharma I&I co-development partnership or a massive secondary equity offering / $200M ATM drawdown) to fund Phase 3 trials in AD and Asthma.
7. Grey Thinking, Conditional Synthesis & The Forensic Verdict
Rejecting both uncritical retail cheerleading and reflexive bear dismissal, we synthesize our findings into five decision-grade “If-Clauses” followed by our explicit, probability-weighted Forensic Verdict.
Decision-Grade “If-Clauses”
-
IF Angel Pharmaceuticals’ China Phase 1b/2 AD trial (Cohort 1 readout expected late 2026) demonstrates continued EASI-75 deepening between Week 8 and Week 12 without plateauing or liver/infectious toxicity,
THEN the primary duration risk going into the 200-patient US Phase 2SIERRA-1trial (12-week endpoint) will be substantially de-risked, likely triggering a pre-readout institutional re-rating of CRVS back toward the $14.00–$17.00 range. -
IF the Phase 3 PTCL trial (
NCT06561048) passes its early 2027 interim futility analysis and maintains an ORR ≥ 32% and median PFS ≥ 5.5 months against belinostat/pralatrexate (expected ~2.5–3.5 months PFS),
THEN Dr. Miller’s prospective 1-to-3 prior-line inclusion filter will be clinically vindicated, establishing a standalone $400M–$600M commercial oncology floor (via Fast Track / Orphan Drug registration) that insulates Corvus’s valuation even before the immunology franchise matures. -
IF Phase 2
SIERRA-1(NCT07441395, mid-2027) delivers a Week 12 placebo-adjusted EASI-75 delta ≥ 32% (e.g., ≥ 55% active vs. ≤ 23% placebo) and vIGA 0/1 delta ≥ 25% in the 200 mg BID arm with sustained 90-day off-treatment Treg remission,
THEN soquelitinib will be validated as a first-in-class oral immune-rebalancing therapy on par withKT-621anddupilumab. In this scenario,@HOThomasWPhelps’s “Not Now” thesis will be completely vindicated, enabling Corvus to command a $1.0B+ upfront / multi-billion-dollar biobucks Big Pharma I&I partnership (or $3.0B+ M&A buyout) at $28.00–$38.00+ per share. -
IF the 400 mg once-daily (QD) arm in
SIERRA-1exhibits lower EASI-75 efficacy or higher lymphopenia/infection rates than the 200 mg twice-daily (BID) arm,
THEN our biophysical RLK covalent spillover equation (IC50,RLK = 261 nM) will be empirically confirmed in humans: highCmaxbolus dosing breaches the 115-fold ITK/RLK kinetic buffer and suppresses Th1/Treg plasticity, locking soquelitinib into an obligate twice-daily (BID) dosing profile (or requiring a modified-release ER formulation for Phase 3). -
IF
SIERRA-1suffers classic Phase 1-to-Phase 2 multicenter compression—landing at a placebo-adjusted EASI-75 delta of 12%–18% due to a 35%+ placebo response across 200 patients—OR if any EBV/opportunistic viral reactivation signal emerges in long-duration cohorts,
THEN soquelitinib will be viewed as commercially non-competitive against once-daily oralKT-621(STAT6 degrader) anddupilumab, compressing CRVS shares back toward $3.50–$5.50 (cash plus PTCL stub value) and forcing dilutive reliance on the $200M Jefferies ATM facility.
The Forensic Verdict: What We Actually Think
After cross-examining every SEC filing, clinical protocol, and enzymatic assay, we arrive at a nuanced but unambiguous verdict on @HOThomasWPhelps’s thesis and Corvus Pharmaceuticals at $9.84 per share:

Probability-Weighted Expected Value: Weighting our three scenarios (
0.50 × $19.00 + 0.25 × $33.00 + 0.25 × $4.50) yields a 12-month probability-weighted expected value of $18.88 per diluted share (+91.9% upside from $9.84), representing an attractively skewed risk/reward profile after the stock’s 63.5% drawdown from its January 2026 highs. -
Where We Agree with
@HOThomasWPhelps:- Biochemical Differentiation is Real: Unlike the failed reversible or dual ITK/RLK inhibitors in the pharma graveyard (
PRN694,PF-06465469,BMS-509744), soquelitinib’s 115-fold kinetic selectivity (kinact/KI) for ITK Cys442 over RLK and its scRNA-seq-verified BACH2/RORγt Th17-to-Treg switch represent genuine, peer-reviewed molecular innovation. - Dr. Richard Miller’s PTCL Phase 3 Design is Astute: Restricting the 150-patient Phase 3 PTCL trial (
NCT06561048) to 1–3 prior lines directly capitalizes on the Phase 1 finding that immune-mediated lymphoma clearance requires an intact host lymphocyte compartment. - “Not Now” at $9.84 is Correct Corporate Finance: With ~$199M in estimated Q3 cash (funded at $22.15/share in January 2026) and four catalysts over the next 9 months, signing away global I&I rights at a depressed ~$700M enterprise value today would be a major capital-allocation error.

Where Investors Must Apply Strict Forensic Discipline:
- Discard the ICB Combo Hype: Do not assign current valuation weight to human anti-PD-1/anti-CTLA-4 combinations; Corvus’s clinical RCC ICB trial was with
ciforadenant(which missed statistical significance and was shelved), and soquelitinib + ICB remains strictly preclinical. - Respect the Cohort 4 Small-Sample CI and Prior-Systemic Skew: Do not extrapolate a 75% EASI-75 rate from
n = 12patients (95% CI: 42.8%–94.5%) linearly into a 200-patient multicenter trial without accounting for the 58% vs. 42% prior-systemic placebo imbalance in Cohort 4. - Watch the 200 mg BID vs. 400 mg QD Arm in
SIERRA-1and Post-Data Partnering: Because of the RLK spillover kinetics at highCmax, 200 mg BID remains the biologically superior regimen. And while “Not Now” is the right answer in October 2026, Corvus must partner its global I&I franchise (or Asthma/ex-US rights) immediately after the mid-2027SIERRA-1readout rather than attempting to self-fund a $600M+ Phase 3 AD/Asthma campaign via its $200M Jefferies ATM shelf.
- Discard the ICB Combo Hype: Do not assign current valuation weight to human anti-PD-1/anti-CTLA-4 combinations; Corvus’s clinical RCC ICB trial was with
