Six days ago, in our October 8, 2026 forensic investigation—The $40B Oncology Paradox: A Forensic Deep Dive into Revolution Medicines ($RVMD), the FDA Review, and the G12D Efficacy Chasm—we documented a critical structural crack in the precision oncology sector’s most crowded consensus trade.
While Wall Street priced Revolution Medicines (NASDAQ: RVMD) at a $40.3 billion to $41.6 billion market capitalization on the premise that its newly approved multi-RAS(ON) inhibitor Rasonque (daraxonrasib / RMC-6236) would serve as an unassailable universal monotherapy across pancreatic ductal adenocarcinoma (PDAC), the U.S. Food and Drug Administration’s unredacted CDER Multidiscipline Review of the Phase 3 RASolute 302 trial (N=228) revealed an acute bimodal split. Whereas daraxonrasib achieved a 50% confirmed objective response rate (cORR) in KRAS G12V (~31% of PDAC), efficacy in KRAS G12D—the dominant mutation driving ~40% to 43% of all pancreatic cancers—dropped to 24% cORR (23/95). We demonstrated that this 26-percentage-point chasm is governed by hard structural biophysics: the negatively charged Asp12 (-CH₂-COO⁻) carboxylate side chain introduces electrostatic repulsion and steric hindrance against the Cyclophilin A (CypA) immunophilin chaperone required by daraxonrasib’s tri-complex mechanism, while off-tumor wild-type RAS (KRAS/NRAS/HRAS) inhibition caps dosing at 300 mg QD due to mucosal ulceration, stomatitis, and rash.
On the morning of October 14, 2026, the competitive consequence of that exact G12D vulnerability materialized in an official SEC Form 8-K filing (Accession No. 0001104659-26-115687) and press release from Verastem Oncology (NASDAQ: VSTM).
Reporting its first multi-tumor U.S. clinical dataset from the Phase 1/2 TARGET-D 101 trial (NCT07020221) for VS-7375 (GFH375)—an oral, non-chaperone, direct KRAS G12D dual ON/OFF inhibitor licensed from Shanghai-based GenFleet Therapeutics—Verastem placed a prominent dark-blue callout box directly onto Slide 12 of its SEC-furnished investor presentation: “Daraxonrasib KRAS G12D: 24% cORR (23/95)”, contrasting RVMD’s subgroup performance against VS-7375’s 50% cORR (4/8) in 2L PDAC and 41% cORR (9/22) in 2L+ PDAC (including two complete responses [CRs]) at 900 mg once daily (QD):

Simultaneously, Verastem executed a classic triple-catalyst corporate maneuver before the opening bell:
- Clinical Data & Phase 3 Alignment: Disclosed multi-tumor efficacy across PDAC, NSCLC, and CRC alongside FDA protocol alignment on a three-arm Phase 3 first-line mPDAC trial (TARGET-D 301,
N=354) featuring a chemotherapy-free arm (VS-7375 900 mg QD + anti-EGFR mAb). - Massive Balance Sheet Dilution: Priced an underwritten public offering of 30,937,500 common shares at $8.00 per share plus 312,503 pre-funded warrants at $7.9999 (led by Jefferies, Guggenheim Securities, Cantor Fitzgerald, and B. Riley) to raise $250 million in gross proceeds—a +7.4% premium to its October 13 closing price of $7.45 (which had tumbled
-9.70%during the prior session as wall-crossed institutions positioned for the bookbuild). - C-Suite Succession: Announced that longtime President and CEO Daniel W. Paterson will retire effective January 1, 2027, handing leadership to Board member and former AVEO Pharmaceuticals CEO Michael P. Bailey.
Across biotechnology desks, the data drop immediately ignited a three-way valuation and scientific debate linking Verastem ($VSTM, ~$675M pre-deal / ~$925M post-deal market cap), Erasca ($ERAS, ~$5.4B–$5.6B market cap at $14.78), and Revolution Medicines ($RVMD, ~$40.5B market cap at ~$188):

“So VSTM is the new RVMD ?”
— Hazem Samara (@hazemsamara), Market Commentator, October 14, 2026
“Why didn’t $VSTM submit an abstract for ESMO to present its G12D data?”
— Adam Feuerstein (@adamfeuerstein), Senior Biotech Journalist, October 14, 2026
“$VSTM - CRC data intriguing but too low N to excite. Clearly have a drug in PDAC & NSCLC, but will take time & execution. 2% discontinuation = tolerability edge (which mgmt contends allowed them to dose higher and achieve better saturation, ultimately better efficacy… TBD)”
— Jonathan Faison (@jfais20), Biotech Analyst, October 14, 2026
Rather than accepting corporate slide decks at face value or collapsing into binary cheerleading, this investigation applies our 8-phase “Panning for Gold” critical thinking framework across the entire RAS oncology triad ($VSTM, $ERAS, $RVMD). We dissect why VS-7375’s direct salt-bridge chemistry corroborates our October 8 biophysical critique of $RVMD, expose the buried U.S. 600 mg denominators (4/27 = 14.8% cORR in PDAC; 3/24 = 12.5% cORR in CRC) that Verastem omitted as percentages in its press release text, answer Adam Feuerstein’s ESMO 2026 question using clinical cutoff math, interrogate why Erasca ($ERAS) trades at an 8x valuation multiple over $VSTM despite a tiny N=7 PDAC denominator and an active U.S. Patent No. 12,409,225 / trade secret dispute with $RVMD, and synthesize decision-grade If-Clauses into an explicit institutional verdict.

1. Phase 1: Issue Classification, Boundary Framing & Falsifiability Criteria
Following Chapter 2 of Browne & Keeley’s Asking the Right Questions, we separate our inquiry into explicit Descriptive (mechanistic/statistical) and Prescriptive (capital allocation/valuation) questions, anchored by strict falsifiability criteria.
Core Investigative Questions
- Descriptive (Biophysical & Clinical): Does Verastem’s VS-7375 overcome the Cyclophilin A steric/electrostatic potency ceiling that restricted Revolution Medicines’ daraxonrasib to a 24% cORR (
23/95) in KRAS G12D PDAC—and why did VS-7375 require a 50% dose escalation from 600 mg QD (the Chinese RP2D) to 900 mg QD in U.S. patients to achieve a 41% confirmed ORR (9/22)? - Descriptive (Regulatory & Statistical): What do the exact binomial confidence intervals, buried 600 mg cohort denominators (
4/27in PDAC,3/24in CRC), ESMO 2026 abstract timelines, and Phase 3 TARGET-D 301 crossover rules reveal about the statistical maturity and regulatory risk of VS-7375? - Prescriptive (Relative Valuation Across
$VSTM,$ERAS,$RVMD): In light of Verastem’s October 14 data and $250M financing at $8.00/share, how should institutional portfolios weight Verastem (~$925M post-deal market cap), Erasca (~$5.5B market cap), and Revolution Medicines (~$40.5B market cap)?
Explicit Falsifiability Criteria
What specific empirical evidence would prove our thesis wrong across each leg of the triad?
- For Verastem (
$VSTMBullishness Falsified): If the Phase 2 registration-directed TARGET-D 201 trial (NCT07644559,N=80–120in 2L KRAS G12D PDAC at 900 mg QD, readout Q2 2027) regresses from the Phase 1/2 41%–50% cORR down to ≤ 25% cORR by blinded independent central review (BICR), or if cumulative exposure beyond 6 months at 900 mg QD triggers late-onset hepatotoxicity or >15% treatment discontinuations. - For Revolution Medicines (
$RVMDSkepticism Falsified): If the Phase 3 RASolute 309 doublet (daraxonrasib + zoldonrasib, NCT06915220) or frontlinezoldonrasib + chemo(RASolute 305) demonstrates >55% confirmed ORR in G12D PDAC with <10% discontinuations and successfully blocks community oncologists from switching G12D patients to direct non-chaperone oral G12D inhibitors (VS-7375, Incyte’sINCB161734). - For Erasca (
$ERASSkepticism Falsified): IfERAS-0015maintains a confirmed ORR ≥ 45% across ≥ 50 specifically KRAS G12D-mutated U.S. PDAC patients inAURORAS-1(NCT06983743) and Erasca secures a declaratory judgment invalidating or proving non-infringement of Revolution Medicines’ U.S. Patent No. 12,409,225 and dismissing all trade secret claims.

Conflicts of Interest & Capital Markets Context
- Verastem’s Wall-Crossed Offering Architecture: On October 14, 2026, Verastem’s clinical data release was synchronized to the minute with a $250 million underwritten equity offering managed by Jefferies, Guggenheim Securities, Cantor Fitzgerald, and B. Riley. Concurrent with the offering, Verastem terminated its August 2025 at-the-market (ATM) prospectus with Cantor Fitzgerald (
0 shares sold). The slide deck (Exhibit 99.3) was explicitly constructed as a marketing document to clear a $250 million bookbuild equal to ~37% of the company’s prior-day market cap. - Erasca’s $891M 2026 Equity Machine & Litigation Overhang: Erasca raised $242.7 million net in January 2026 and another $632.5 million gross in July 2026 (~$891.3 million total 2026 gross offerings) while facing active shareholder class action lawsuits triggered by its April 27, 2026 disclosure that Revolution Medicines accused
ERAS-0015of infringing U.S. Patent No. 12,409,225 and misappropriating trade secrets.
3. Phase 3: Semantic Precision & The Operational Substitution Test
Biotechnology investor presentations rely heavily on elastic terminology. We isolate three central claims from today’s RAS disclosures and subject them to the Browne & Keeley Substitution Test:

1. “Emerging Best-in-Class RAS Inhibitor in 2L PDAC (50% cORR)” ($VSTM)
- Definition A (Strict / Registration-Grade): A statistically powered, Blinded Independent Central Review (BICR) confirmed objective response rate across N ≥ 80 second-line PDAC patients whose lower 95% confidence bound exceeds competing benchmarks (>30%).
- Definition B (Loose / Exploratory Subgroup Point Estimate): An investigator-assessed point estimate of 50% (
4/8patients) within a 22-patient Phase 1/2 expansion cohort at a newly escalated 900 mg QD dose. - The Substitution Impact: Under Definition B, Verastem’s claim is factually accurate and supported by deep biomarker clearance (
100%of evaluable patients had ≥ 50% ctDNA VAF reduction;58%had100%clearance) and two complete responses in 3L/4L disease. Under Definition A, however, $4/8$ carries an exact 95% Clopper-Pearson confidence interval of 15.7% to 84.3%—meaning a single patient shifting from PR to SD moves the 2L headline from 50% to 37.5%.
2. “57% uORR (8-week) in 2L+ KRAS G12X PDAC” ($ERAS)
- Definition A (Strict / Subtype-Verified Confirmed Efficacy): Confirmed RECIST v1.1 responses (≥ 2 consecutive scans ≥ 4 weeks apart) specifically within the hard-to-treat KRAS G12D subpopulation across a meaningful sample size (N ≥ 30).
- Definition B (Loose / Early Unconfirmed Pan-Mutant Cut): Combining unconfirmed single-scan shrinkages at 8 weeks across all KRAS G12X mutations (lumping high-responder G12V together with G12D) in a tiny cohort of $N = 7$ patients (
4/7) at 32 mg QD. - The Substitution Impact: Wall Street currently values Erasca at ~$5.5 billion as though
ERAS-0015has satisfied Definition A. In reality,ERAS-0015has only satisfied Definition B (4/7unconfirmed responses pooled across G12X subtypes). As we proved with$RVMD, pooling G12V (50% cORR) and G12D (24% cORR) in a CypA molecular glue creates a severe aggregation illusion.
3. “FDA Alignment on a Chemo-Free Frontline Phase 3 PDAC Regimen” ($VSTM)
- Definition A (Strict / De-Risked Registrational Path): FDA agreement on a Phase 3 protocol after reviewing robust Phase 2 randomized efficacy and survival data for the exact experimental combination in the target tumor type.
- Definition B (Loose / Protocol Clearance): FDA regulatory permission to include an experimental chemo-free arm (
VS-7375 900 mg QD + anti-EGFR mAb,N=118) alongside aVS-7375 + GnParm (N=118) and chemotherapy control (N=118) in Phase 3 TARGET-D 301 before initial Phase 1/2VS-7375 + cetuximabPDAC efficacy data have even been reported (slated for year-end 2026). - The Substitution Impact: While FDA alignment (
Definition B) is a major regulatory milestone that accelerates Verastem’s timeline into 1L PDAC by year-end 2026, investors must recognize that Arm A (VS-7375 + anti-EGFRwithout chemotherapy in frontline metastatic pancreatic cancer) is a high-variance clinical bet, not a de-risked certainty.
4. Phase 4: Biophysical & Strategic Disentanglement (4-Way Causal Matrices)
1. Why Does VS-7375 Succeed in G12D Where Daraxonrasib Stumbles?
To understand how today’s Verastem data directly validates our October 8 deep dive into Revolution Medicines, compare the molecular architectures of the two classes:

In short: the exact chemical feature that cripples daraxonrasib in KRAS G12D—the negatively charged Asp12 carboxylate group—is the primary binding anchor that gives VS-7375 its selectivity and therapeutic window. Because VS-7375 does not touch wild-type KRAS, NRAS, or HRAS in healthy human epithelium (Slide 30: 0% mucositis, 0% stomatitis, 0% Grade ≥ 3 rash across N=48 patients at 900 mg QD), Verastem can push systemic exposure into the deep target-saturation zone without hitting a wild-type toxicity wall.
2. Why Did 600 mg QD Work in China (GFH375) But Collapse to 14.8% cORR in the U.S. (VS-7375)?
One of the most striking revelations buried in today’s filing is the divergence between 600 mg QD and 900 mg QD in U.S. patients. We test the 4-way causal matrix:

Look closely at Slide 15 from Verastem’s SEC Form 8-K (Exhibit 99.3) below:

In the press release text, Verastem wrote: “Compared with 600 mg QD monotherapy (n=27 mITT), the 900 mg dose produced deeper tumor reductions and a higher response rate. At 600 mg, eight patients achieved objective responses, including four confirmed responses, and 11 of 27 patients remain on treatment.” Notice why Verastem omitted the percentage: $4 / 27$ is a 14.8% confirmed ORR (and 8/27 = 29.6% unconfirmed ORR), with 59% (16/27) of patients already off treatment!
Why did 600 mg QD produce a 41% ORR in GenFleet’s Chinese trial (GFH375, ESMO 2025 Abstract LBA84) but only 14.8% cORR (4/27) in Verastem’s U.S. trial?
- Because oral small-molecule volume of distribution (V_d) and hepatic clearance scale with body weight and body surface area. In a U.S. oncology population (
82% White, median body weight typically 20%–30% higher than East Asian cohorts), a flat 600 mg QD dose diluted plasma AUC_ss down to the middle green box on Slide 15 (N=59), leaving more than half of U.S. patients below the preclinical target required for universal tumor regression. - Escalating by 50% to 900 mg QD (
N=23PK) shifted the human AUC_ss distribution up into the purple box—above the 100 mg/kg murine equivalent threshold—resulting in zero target lesion growth across all 20 evaluable waterfall patients, 9 confirmed responses (41% cORRin 2L+,50% cORRin 2L), and two complete responses (-100%SLD) in 3L and 4L PDAC!
3. Answering Adam Feuerstein: Why Didn’t $VSTM Submit an ESMO 2026 Abstract?
On X this morning, senior biotech journalist Adam Feuerstein (@adamfeuerstein) posed a pointed question: “Why didn’t $VSTM submit an abstract for ESMO to present its G12D data?”
Applying temporal and causal auditing reveals three concrete reasons—none of which require conspiracy theories, but all of which highlight the tension between academic congress schedules and biotech capital raises:
- The ESMO Abstract Deadline vs. The 600 mg Trap: The ESMO Congress 2026 (taking place October 23–27, 2026 in Madrid) closed its regular abstract submission window in May 2026 (and Late-Breaking Abstract intent declarations in early summer). At that time, Verastem’s U.S.
TARGET-D 101dataset consisted almost entirely of the 600 mg QD cohort—where U.S. 2L+ PDAC cORR was sitting at a dismal 14.8% (4/27) and U.S. 3L+ mCRC + cetuximab cORR was sitting at 12.5% (3/24)! Meanwhile, the 900 mg QD mITT cutoff wasn’t even locked until July 22, 2026 (requiring follow-up through October 7, 2026 to confirm the 9 responses on second scans). Submitting a spring/summer ESMO abstract on the U.S. trial would have forced Verastem to publish a placeholder abstract dominated by under-dosed 600 mg U.S. patients. - GenFleet Already Presented
GFH375Monotherapy at ESMO 2025 (LBA84) and Focused ESMO 2026 onGFH276: Because GenFleet had already headlinedGFH375PDAC monotherapy in a late-breaking oral session at ESMO 2025 (Abstract LBA84), ESMO committees rarely grant a second late-breaking oral slot to the same Phase 1/2 molecule merely for a Western bridging dose-escalation from 600 mg to 900 mg (meanwhile, GenFleet securedLBA33at ESMO 2026 for its next-generation pan-RAS inhibitorGFH276, while Incyte secured Rapid Oral slots#1007ROforINCB161734). - Synchronizing the $250M Wall-Crossed Offering & CEO Transition: Had Verastem submitted to ESMO 2026, it would have been bound by strict ESMO embargo rules until late October, preventing management from selectively showing the October 7 data cutoff to wall-crossed institutional investors in mid-October to price a $250 million underwritten offering at $8.00/share alongside CEO Daniel Paterson’s retirement announcement.
5. Phase 5: Quantitative Audit, Statistical Confidence Intervals & Capital Math
Numbers in early-phase oncology press releases create an illusion of precision when small denominators are converted into bold percentages. We perform a rigorous statistical audit across all competing KRAS G12D datasets and balance sheets.
1. Exact Binomial Confidence Interval Audit (Clopper-Pearson 95% CIs)
Let us strip away the bar-chart heights on Verastem’s Slide 12 and Erasca’s press releases and calculate the exact 95% Clopper-Pearson binomial confidence intervals for every reported response rate in previously treated PDAC:
$$\text{95% Exact Clopper-Pearson CI for } \hat{p} = \frac{k}{n}: \quad \left[ \text{Beta}^{-1}\left(0.025; k, n-k+1\right), ; \text{Beta}^{-1}\left(0.975; k+1, n-k\right) \right]$$

2. What the Blinding Test Reveals Across Tumor Types
When we inspect the raw numbers across all cohorts in Verastem’s TARGET-D 101 update without promotional adjectives, three distinct clinical realities emerge:
- In mPDAC (900 mg QD Monotherapy,
n=22mITT): Even accounting for small-$N$ variance (95% CI [20.7%, 63.6%]), the combination of 9/22 (41%) confirmed responses (which is 9/20 = 45% among evaluable patients who reached their first scan), two complete responses (-100%target lesion disappearance in 3L and 4L PDAC), 88% (15/17) achieving >50% CA19-9/CEA reductions, and 100% (12/12) achieving >50% KRAS G12D ctDNA VAF reductions (with58% [7/12]achieving 100% ctDNA clearance) is an unmistakable, high-conviction biological signal. Complete responses in 3L/4L metastatic pancreatic cancer with an oral monotherapy are exceedingly rare. - In Advanced NSCLC (600 mg QD Monotherapy,
n=23mITT; + 1L Combon=8): Why did 600 mg QD achieve a 43% confirmed ORR (10/23) and 100% (5/5) complete ctDNA VAF clearance in 2L+ NSCLC (Slide 20–22), whereas 600 mg QD struggled in U.S. PDAC (14.8% cORR) and CRC (12.5% cORR)?- Mechanistic Tissue Perfusion Reason: Pancreatic tumors are characterized by dense, hypovascular desmoplastic stroma that impairs drug penetration, requiring higher plasma AUC_ss (900 mg QD) to saturate intracellular KRAS G12D. Lung adenocarcinomas are highly vascularized, allowing 600 mg QD to achieve 100% (
5/5) ctDNA clearance and a 43% cORR (even with 30% of patients having failed prior taxanes) and a 72% 6-month PFS rate! Moving NSCLC to 900 mg QD in Phase 2 TARGET-D 202 (NCT07659782) provides further upside.
- Mechanistic Tissue Perfusion Reason: Pancreatic tumors are characterized by dense, hypovascular desmoplastic stroma that impairs drug penetration, requiring higher plasma AUC_ss (900 mg QD) to saturate intracellular KRAS G12D. Lung adenocarcinomas are highly vascularized, allowing 600 mg QD to achieve 100% (
- In 3L+ mCRC (Combination with Cetuximab): As Jonathan Faison (
@jfais20) rightly noted, the colorectal data are currently too low-$N$ to declare victory. At 600 mg QD + cetuximab (n=24mITT), confirmed ORR was only 12.5% (3/24 cPR,1 uPR). At 900 mg QD + cetuximab (n=6mITT,5evaluable), Verastem reported 33% ORR (2/6), but only 1 of those 2 responses is confirmed (1/6 = 16.7% cORR,1 uPR). While 5 of 6 patients remain on therapy with zero target lesion growth (100% DCRin evaluable), CRC requires the mature year-end 2026 readout before assigning material commercial value.

Look at the Enterprise Value (EV) asymmetry revealed by this table:
- Verastem (
$VSTM)—which already generates$24.0 million per quarter ($96M/year annualized) in commercial net revenue from its FDA-approved ovarian cancer therapy AVMAPKI FAKZYNJA CO-PACK (with a Blister Pack NDA just granted FDA Priority Review on October 7, 2026 for a Q1 2027 decision) AND holds ex-China rights to VS-7375 (now entering Phase 3 1L PDAC with41% cORRin 2L+ PDAC and43% cORRin 2L+ NSCLC)—trades at a pro-forma Enterprise Value of roughly $720M to $750M! - Erasca (
$ERAS)—which has $0 commercial revenue, onlyN=7unconfirmed 8-week PDAC patients at its 32 mg RDE (4/7across pooled G12X), and a direct patent infringement and trade secret legal threat from Revolution Medicines—trades at an Enterprise Value of ~$4.5B to $4.6B (6x Verastem’s EV)! - Revolution Medicines (
$RVMD) trades at an Enterprise Value of ~$36.8B (~50x Verastem’s EV) while burning $535M per quarter and defending a 24% G12D response rate that Verastem just publicly targeted on Slide 12.
6. Phase 6: The “Negative View” & Omission Interrogation
Persuasive biotech pitches highlight best-case responder bars while burying structural liabilities. We interrogate the omitted risks across all three companies:
1. What Could Go Wrong for Verastem ($VSTM)?
- The Phase 3
TARGET-D 301Crossover & Chemo-Free Design Risks: Look closely at Slide 36 of Verastem’s SEC Form 8-K presentation (Exhibit 99.3):

There are two major structural risks embedded in this 354-patient (118/arm) trial schema:
- Control Arm Crossover Confounds Overall Survival (OS): The protocol explicitly states: “SoC (GnP or FOLFIRINOX) arm crossover to VS-7375 monoTx (or with α-EGFR) allowed after progression.” While crossover is ethically attractive and will help
TARGET-D 301enroll rapidly against competing frontline trials (RASolute 305, Incyte’sDAWN-303, Astellas’ASP3082), allowing control patients to cross over to activeVS-7375after progression will systematically dilute the secondary Overall Survival (OS) hazard ratio. If the FDA demands a clean OS trend for full frontline approval, crossover could complicate regulatory review. - Unproven Frontline Chemo-Free Arm (
Arm A): Randomizing 118 treatment-naïve metastatic pancreatic cancer patients to a chemo-free regimen (VS-7375 900 mg + anti-EGFR Ab) head-to-head againstmFOLFIRINOX(mPFS ~6.4 mo) orGnP(mPFS ~5.5 mo) before Verastem has even reported Phase 1/2VS-7375 + cetuximabdata in PDAC is a bold gamble. Fortunately, the 3-arm design includes Arm B (VS-7375 900 mg + GnP) as an independent shot on goal.
- Short Follow-Up Duration at 900 mg QD (
mDoT = 3.0 months): In the safety tables (Slides 27, 45, 52), Verastem emphasizes that Grade ≥ 3 TRAEs were lower at 900 mg QD (10%,N=48) than at 600 mg QD (19%,N=73), and dose interruptions were15%vs21%. However, notice the confounder on Slide 45: median duration of treatment (mDoT) at 900 mg QD was only 3.0 months (range 0.2–10.6) compared to 4.3 months at 600 mg QD. Furthermore, in the 1L NSCLC triplet combination (VS-7375 600 mg + carboplatin/pemetrexed/pembrolizumab,N=9,Slide 48), Grade 3 diarrhea reached 22% (2/9) and Grade 3/4 neutropenia reached 22% (2/9). Escalating the 1L NSCLC triplet to 900 mg QD will require close monitoring of overlapping GI and hepatic toxicities. - GenFleet Territorial Split & Milestone Encumbrances:
Unlike a wholly owned asset, Verastem holds rights to
VS-7375outside Greater China (GenFleet retains mainland China, Hong Kong, Macau, and Taiwan) and owes GenFleet up to $625.5 million in total development, regulatory, and commercial milestones across their 3-program collaboration plus tiered royalties on net sales, on top of Verastem’s Oberland royalty financing facility.
2. What Could Go Wrong for Erasca ($ERAS)?
- The Revolution Medicines Patent ’225 & Trade Secret Time-Bomb:
In its Q2 2026 Form 10-Q and July 13, 2026 release, Erasca explicitly warned investors of:
“…our ability to successfully defend against allegations raised by, or any litigation initiated by, Revolution Medicines (RevMed) that ERAS-0015 infringes patents held by RevMed or was derived from RevMed trade secrets.” On April 24, 2026, Revolution Medicines formally notified Erasca that
ERAS-0015infringes U.S. Patent No. 12,409,225 under the doctrine of equivalents and alleged trade secret misappropriation tied toERAS-0015’s origin (licensed from Guangzhou Joyo Pharmatech, to whom Erasca paid a massive $150.0 million IPR&D fee in Q1 2026 to expand rights to Greater China). - The CypA Glue Paradox:
Even if Erasca prevails in court after years of costly patent litigation,
ERAS-0015is a pan-RAS Cyclophilin A molecular glue. If its binding mode to active RAS(ON) via CypA mirrorsdaraxonrasib, what happens whenERAS-0015expands fromN=7pooled KRAS G12X PDAC patients (57% uORR_8wk) to a 100-patient Phase 3 cohort where ~43% of patients carry the negatively charged KRAS G12D mutation? At a ~$5.5 billion market cap,$ERASis priced for perfection before answering either its G12D subtype question or its Patent ’225 litigation threat.
3. How This Relates to Revolution Medicines ($RVMD)
Today’s Verastem disclosure is the empirical smoking gun for the thesis we laid out in our October 8 $RVMD deep dive:
- The End of the “Unassailable G12D Monopoly” Premium: On October 8, sell-side analysts defending their unanimous 20-out-of-20 “Buy” ratings on
$RVMDargued that a 24% cORR (23/95) fordaraxonrasibin 2L KRAS G12D PDAC didn’t matter because no competing oral agent had demonstrated superior confirmed responses and tolerability in Western G12D PDAC patients. Six days later, Verastem put 41% cORR (9/22) in 2L+, 50% cORR (4/8) in 2L, two complete responses, 0% mucositis/stomatitis, and 2% discontinuations onto an SEC Form 8-K slide right next toDaraxonrasib KRAS G12D: 24% cORR (23/95). - RVMD’s Doublet Crutch vs. Single-Agent Oral Selectivity: Because
daraxonrasib(24% cORR in G12D) andzoldonrasib / RMC-9805(only 15% confirmed ORR [6/40] / 30% uORR in 2L+ G12D PDAC per Slide 12 Footnote 1!) each have modest confirmed monotherapy response rates in G12D PDAC, Revolution Medicines was forced to combine both CypA glues in Phase 3 RASolute 309 (NCT06915220, initiated October 5, 2026). By contrast, a direct dual ON/OFF inhibitor likeVS-7375achieves deep G12D pathway shutdown (100%of patients with >50% ctDNA VAF drop) as a single oral agent, leaving its therapeutic window wide open to combine cleanly with standard-of-careGnP,anti-EGFR, orchemo/pembrolizumab.
7. Phase 7: Grey Thinking, Conditional Synthesis & The Forensic Verdict
Rejecting binary tribalism, we structure our strategic conclusions through explicit If-Clauses before rendering our probability-weighted institutional verdict across $VSTM, $ERAS, and $RVMD.
Decision-Grade Conditional “If-Clauses”
- IF Verastem’s Phase 2 registration-directed
TARGET-D 201trial (NCT07644559, 2L KRAS G12D PDAC at 900 mg QD, readout Q2 2027) confirms a BICR response rate ≥ 35% with median PFS ≥ 6.5 months and <5% AE discontinuations,
THENVS-7375will have an open regulatory lane for FDA Accelerated Approval in 2L KRAS G12D PDAC, directly displacingdaraxonrasibin the largest genetic subset of pancreatic cancer (~43% of patients) and warranting a re-rating of$VSTMfrom ~$1.0B post-deal market cap toward $2.5B–$3.5B ($19–$26/share). - IF
TARGET-D 201BICR confirmed ORR regresses into the 22%–26% range once sample size expands from $N=22$ to N ≥ 80 (mirroring the historical phase-transition decay seen in Incyte’sINCB161734confirmed vs. unconfirmed split),
THENVS-7375’s differentiation overdaraxonrasibin 2L PDAC will narrow to tolerability (0% mucositis/stomatitis vs. pan-RAS toxicity) rather than superior tumor shrinkage, anchoring$VSTMnear $6.00–$8.50/share (supported by its ~$455M pro-forma cash and ~$96M/yearAVMAPKI FAKZYNJAcommercial franchise). - IF Erasca’s
ERAS-0015shows a sharp efficacy drop-off between KRAS G12V and KRAS G12D whenAURORAS-1(NCT06983743) reports larger subtype-stratified confirmed ORR in H1 2027—or if Revolution Medicines escalates its April 2026U.S. Patent No. 12,409,225notice into a formal federal preliminary injunction suit,
THEN Erasca’s ~$5.5B market valuation ($14.78/share) will suffer a severe multiple compression back toward $2.0B–$2.5B ($5.50–$7.00/share).

1. Stance on the Scientific & Clinical Evidence
$VSTM‘s 900 mg QD Dose Solves the U.S. Exposure Deficit and Validates Direct G12D Selectivity: While we flag the small sample size in 2L PDAC (4/8 = 50% cORR,95% CI [15.7%, 84.3%]) and expose the under-dosed U.S. 600 mg cohort (4/27 = 14.8% cORR), the totality of evidence at 900 mg QD—9/22 (41%) cORRacross 2L–4L PDAC, two complete responses in 3L/4L disease, 100% (12/12) >50% ctDNA VAF reduction (58%complete clearance), 43% cORR (10/23) in 2L+ NSCLC even at 600 mg, 0% mucositis/stomatitis, and a 2% discontinuation rate—demonstrates that direct dual ON/OFF G12D inhibition avoids the CypA steric hindrance and wild-type toxicity ceiling that trapped Revolution Medicines’daraxonrasibat 24% cORR in G12D.$VSTM’s Governance & Financing Execution Was Opportunistic: Management withheld an ESMO 2026 abstract because the U.S. 600 mg cohort was under-exposed in spring 2026, waited until the 900 mg cohort matured to an October 7 cutoff, and used today’s data to clear a $250 million (31.25Mshare/warrant) dilution at $8.00/share while announcing CEO Daniel Paterson’s retirement. Incoming CEO Michael P. Bailey (who successfully launchedFOTIVDAat AVEO and sold the company to LG Chem in 2023) is a seasoned commercial and M&A operator.
2. Actionable Institutional Portfolio Verdict Across the RAS Triad
- Verastem Oncology (
$VSTM— Rating: SPECULATIVE BUY / ACCUMULATE NEAR $7.50–$8.00 OFFERING CLEARING PRICE; 12-Month Target: $14.00–$18.00):
At a pro-forma Enterprise Value of ~$720M–$750M backed by ~$455M in cash and ~$96M/year in commercial LGSOC revenue, the market is assigning less than $400M of net pipeline value toVS-7375. Once the31.25Mnew offering shares finish churning around the $8.00 deal price,$VSTMoffers the most asymmetric risk-adjusted upside in the KRAS G12D space. - Revolution Medicines (
$RVMD— Rating: REITERATE TACTICAL SHORT / UNDERWEIGHT; 12-Month Target: $95.00–$110.00):
Today’s data from$VSTMdirectly reinforces our October 8 short/underweight thesis. At a ~$40.4B market cap and $2.1B+ annual cash burn,$RVMDcan no longer claim an uncontested monopoly across KRAS-mutant PDAC when its 24% G12D monotherapy response rate is being flanked by selective, well-tolerated oral G12D inhibitors (VS-7375,INCB161734) entering Phase 3. - Erasca (
